Overview:
Epithalon, also known as Epitalon or Ala-Glu-Asp-Gly, is a synthetic tetrapeptide corresponding to a naturally occurring peptide epithalamin, produced by the pineal gland. It has gained attention in aging research for its telomerase-activating effects, antioxidant capacity, and potential to extend healthy lifespan. Discovered by Professor Vladimir Khavinson in the 1980s, Epithalon is classified as a geroprotective agent in preclinical settings and is widely studied for its role in epigenetic regulation, neuroendocrine support, and immunomodulation.

  • Mechanism of Action:

Epithalon exerts its biological effects through both direct interaction with DNA expression machinery and indirect hormonal regulation. Key proposed mechanisms include:

    • Activation of telomerase: Stimulates the enzyme responsible for maintaining telomere length in somatic cells, helping to preserve genomic stability.
    • Antioxidant activity: Reduces lipid peroxidation and scavenges free radicals, particularly in brain and liver tissue.
    • Regulation of melatonin: Influences pineal function and circadian rhythm through modulation of melatonin secretion.
    • Epigenetic influence: Alters gene expression profiles related to inflammation, cell cycle, and apoptosis.

Notably, Epithalon has been shown to upregulate antioxidant enzyme expression, stabilize chromatin, and enhance DNA repair efficiency—contributing to its longevity-promoting properties.

  • Preclinical Studies:

    • Lifespan extension: In rodent models, chronic administration of Epithalon led to significant increases in mean and maximum lifespan, especially in senescent animals.
    • Cancer suppression: Demonstrated inhibition of spontaneous tumor formation and metastasis in aged mice.
    • Neuroendocrine balance: Improved function of the pineal gland, thymus, and hypothalamic-pituitary axis in aged subjects.
    • Immune system rejuvenation: Stimulated T-cell and B-cell activity and increased antibody production in aged animal models.
    • Visual and skin health: Shown to prevent retinal degeneration and support dermal fibroblast regeneration in vitro.
