Overview:
MGF (Mechano Growth Factor) is a naturally occurring splice variant of IGF-1 (Insulin-Like Growth Factor-1) that is rapidly expressed in skeletal muscle in response to mechanical overload or damage. It plays a pivotal role in muscle repair, regeneration, and hypertrophy, particularly by activating satellite cells and stimulating muscle fiber growth.
Synthetic MGF, commonly studied as PEG-MGF (pegylated MGF) is designed to mimic the natural peptide while prolonging its half-life, making it useful in muscle physiology research and regenerative medicine.

What is MGF?
-
Mechanism of Action:
MGF’s function centers around its autocrine and paracrine signaling in muscle tissue:
- Splice Variant Activity: MGF differs from IGF-1 in its E-domain sequence, allowing it to act locally and rapidly after muscular stress.
- Satellite Cell Activation: Promotes proliferation of muscle stem cells, which are crucial for tissue repair and hypertrophy.
- Myogenesis and Regeneration: Increases protein synthesis, muscle fiber recruitment, and muscle cell differentiation.
- Neuroprotection and Cardioprotection: Shown to offer protective effects in neurons and cardiac cells under stress or injury.
-
Preclinical and Clinical Studies:
- Muscle Growth and Repair: MGF promotes muscle mass accrual, especially in damaged or atrophied muscles, making it a candidate for sarcopenia or cachexia models.
- Injury Recovery: Enhances tissue healing post-trauma or surgery by accelerating muscle stem cell activation.
- Neuroprotective Effects: Demonstrated ability to prevent apoptosis in neurons and may play a role in motor neuron survival.
- Cardiovascular Support: Improves cardiac function post-infarction and may support vascular repair mechanisms.
