Overview:

Sermorelin is a synthetic peptide analog of growth hormone–releasing hormone (GHRH 1–29), the natural hypothalamic hormone responsible for stimulating the release of endogenous growth hormone (GH) from the anterior pituitary. Sermorelin replicates the function of endogenous GHRH but with greater pharmacological control and therapeutic application, especially in growth hormone deficiency (GHD) and age-related GH decline.

Unlike recombinant GH therapy, Sermorelin promotes the body’s natural pulsatile secretion of GH, reducing the risk of excessive dosing and supporting IGF-1 modulation within physiological ranges.

  • Mechanism of Action:

Sermorelin acts directly on pituitary GHRH receptors to increase the secretion of endogenous GH. Its effects include:

    • GH Pulse Stimulation: Triggers natural growth hormone pulses, preserving the circadian rhythm of secretion.
    • IGF-1 Elevation: Indirectly elevates insulin-like growth factor 1 (IGF-1) via hepatic stimulation.
    • Muscle & Metabolic Benefits: Supports lean body masslipid metabolism, and tissue repair.
    • Anti-Aging Potential: Addresses age-related GH decline and associated symptoms (fatigue, reduced libido, impaired recovery).

Unlike synthetic GH, Sermorelin does not suppress the hypothalamic-pituitary axis, making it a safer alternative in long-term use.

  • Preclinical and Clinical Studies:

    Pediatric GH Deficiency:

    • Originally FDA-approved for children with idiopathic GHD.
    • Proven to increase height velocity and serum IGF-1 levels.

    Adult GH Deficiency (AGHD):

    • Demonstrated improvements in body compositionenergy, and mood.
    • Shown to normalize IGF-1 levels in middle-aged and elderly adults.

    Anti-Aging and Wellness Therapies:

    • Used in integrative and age-management medicine to address symptoms of somatopause (age-related GH decline).
    • Supports cognitive functionskin elasticity, and recovery.

    Comparative Efficacy:

    • Less risk of edemajoint pain, or glucose intolerance compared to exogenous GH therapy.
    • Often combined with GHRP-2GHRP-6, or Ipamorelin for synergistic GH release.
